学术成果及影响

致力于T淋巴细胞早期发育、T淋巴细胞抗病毒感染免疫以及肿瘤免疫等方面的研究,以第一或通讯作者(含共)在Nat ImmunolCell Stem CellNat CommunPNASJ Biol ChemJ Immunol等高水平学术期刊发表多篇研究性论文。部分代表性论文简介如下:一、发现DNA序列特异性转录因子Tcf1/Lef1具有组蛋白去乙酰化酶(HDAC)活性,直接修饰组蛋白,调控胸腺CD8+ T淋巴细胞早期发育命运决定。研究工作发表于Nature Immunology2016),该研究领域知名专家Dan R Littman(美国科学院院士)在Nature Immunology上同期发表专题评论。二、结合Tcf1调控基因网络和生物信息数据库Connectivity MapCMAP)分析,以及体内外生物学功能筛选,发现小分子化合物PGE1及其FDA批准的PGE1类似物Misoprostol能够阻断Tcf1转录激活的靶基因,治疗白血病(老药物新用途)。该研究工作以封面文章发表于Cell Stem Cell2017),引起了HematologyTimesCBS News以及当地媒体The Daily Iowan等积极追踪报道。三、病毒感染及其多肽疫苗免疫情况下,在TFH细胞发育过程中,转录因子Tcf1通过招募磷酸化pS21 Ezh2作为其转录调控Co-activator,激活TFH细胞发育关键基因Program,促进TFH细胞早期发育;同时通过其自身HDAC活性抑制T细胞共抑制通路(T cell co-inhibitory pathways),保护TFH细胞生物学功能发挥。论文分别发表于Nature Communications2018)、PNAS2021)。多项研究工作被国际权威学术机构F1000收录,以及被Nat Rev ImmunolNat ImmunolImmunityBloodLeukemia等杂志作为亮点推荐。
